Major Accomplishments

A number of important viruses infecting crop plants have been identified and characterized by biological, serological and molecular methods. Notable among them are Groundnut bud necrosis virus (GBNV), Tobacco streak virus (TSV), Papaya ringspot virus, Mungbean yellow mosaic India virus (MYMIV), Tomato leaf curl virus (Different species), Watermelon bud necrosis virus (WBNV), Citrus tristeza virus (CTV), Cardamom bushy dwarf virus, Large cardamom chirke virus (LCCV), Indian citrus ringspot virus (ICRSV), Garlic virus XBanana streak virus (BSV) and Grapevine leafroll associated virus. Diagnostic probes in the form of recombinantly expressed polyclonal antibodies for ELISA, ISEM etc and Primers for PCR and RT-PCR have been developed for major viruses. Transgenic resistance have been developed for major viruses like PRSV, Tomato leaf curl New Delhi virus, GBNV etc and the technology has been transferred to the stake holders.

Referral Centre for virus testing of tissue culture raised plants

Plant Virus Unit is recognized as National referral laboratory for virus diagnostics through a notification dtd. 28th September, 2004 by Dept. Of Biotechnology,  Govt. of India. Under this facility virus testing particularly of tissue culture raised plants is provided and a token fee is levied as per ICAR guidelines.

Objective 1: Development of diagnostic probes/kits for viruses
Objective 2: Development, standardization and validation of virus test protocols
Objective 3: Conformation of results of diagnosis of diagnostics test centers, whenever required
Objective 4: Preparation of standard package of practices and manual 

Expression constructs for serodiagnosis of plant viruses

Traditional method of antiserum production involves using purified virus as antigen. Some of the viruses are difficult to purify and sometimes the host materials present in the virus preparation give high background reaction in serological tests. A number of expressions constructs for the serodiagnosis of plant viruses have been developed at IARI.  These generate protein (antigen) to be used for immunization of rabbit for production of polyclonal antibody. This antibody can be used to prepare different version of kits and products. Expression constructs are available for the following viruses at ACPV.

ELECTRON MICROSCOPY

Morphological identification service through transmission electron microscopy is provided for samples received from different parts of the country and a fee is levied as per ICAR guidelines.

 

MICROARRAY AND CONFOCAL MICROSCOPY
Plant virology unit has recently established a microarray and confocal microscopy facilities to meet the requirements of advances in molecular virology and functional studies of plant virus interaction as well as a supplement to all the host-pathogen interaction studies of the other sections of the Division of Plant Pathology.
Microarray analysis service and Confocal microscopy services will also be provided at the Plant virology unit in near future.
